چکیده
The symbiotic relationships shared between humans and their gastrointestinal parasites present opportunities to discover novel therapies for inflammatory diseases. A prime example of this phenomenon is the interaction roundworms such as hookworm, Necator americanus. Epidemiological observations, animal studies clinical trials using experimental human hookworm infection show that hookworms can suppress inflammation in a safe well-tolerated way, key immunomodulatory properties lies within secreted proteome. Herein we describe identification 2 netrin domain-containing proteins from N. americanus secretome, explore potential treating intestinal mouse models ulcerative colitis. One these proteins, subsequently named Na-AIP-1, was effective at suppressing disease when administered prophylactically acute TNBS-induced model This protective effect validated more robust CD4 T cell transfer chronic colitis, where prophylactic Na-AIP-1 reduced T-cell-dependent type-1 cytokine responses intestine associated pathology. Mechanistic revealed depletion CD11c+ cells abrogated anticolitic Na-AIP-1. A secreted protein from the human hookworm necator americanus binds selectively to NK cells and induces IFN-gamma production.
Parasitic helminths induce chronic infections in their hosts although, with most human helminthiases, protective immunity gradually develops with age or exposure of the host. One exception is infection with the human hookworm, Necator americanus, where virtually no protection ensues over time. Such observations suggest these parasites have developed unique mechanisms to evade host immunity, lea...

Experimental trials are underway exploring the use of parasitic helminthes to treat autoimmune diseases. As with any experimental therapy, such trials must adequately address safety concerns associated with the treatment protocol. In the treatment protocol employed by Autoimmune Therapies, patients are inoculated intradermally with the common nematode hookworm Necator Americanus. Beyond the pot...

Blood-feeding parasites use mechanistically distinct proteases to digest hemoglobin (Hb), often as multienzyme cooperative cascades. We investigated the roles played by 3 distinct proteases from adults of the human hookworm Necator americanus. The aspartic protease Na-APR-1 and the cysteine protease Na-CP-3 were expressed in catalytically active form in yeast, and the metalloprotease Na-MEP-1 w...

The infective larvae of Necator americanus were shown to secrete all mechanistic classes of proteolytic enzymes with two overall pH optima of 6.5 and 8.5 using fluorescein isothiocyanate-labeled casein as the substrate. Since infective larvae are obligate skin penetrators, the effect of each of these enzyme classes against macromolecules derived from human skin was examined. Larval secretions w...
